I couldn’t find anything directly so here are some ideas for you to pursue:
1. I would get a copy of his father Henry’s obituary to see if it listed his family members. You can do that by writing here:

That may or may not help since someone posted at least part of his obituary on his findagrave entry here:

If that’s all there is that won’t be of any help.
2. Henry may have a probate record that listed beneficiaries. Those have been put into a book that’s available at the following libraries. Visit one of them if one is close to you:

5. Are you familiar with the book “William Guffey (1786) ancestors and descendants, and some related families” which can be found in these libraries:
The author is now deceased but he posted this on a message board some years ago.
